Abstract Powdery mildew disease caused by Oidium arachidis poses a threat to peanut production in Africa. Loss of function mutants specific Mlo (Mildew Locus O) genes have provided broad-spectrum and durable resistance against pathogen many crop species. Since there is huge potential utilize susceptibility gene-mediated improvement, genome-wide mining required for further researches. However, the not been characterized genome. In this research study, genome cultivated was used as reference identify AhMlo loci. Our results revealed that 25 loci were identified distributed on chromosomes peanut. Eleven located A-genome while remaining 14 B-genome. Variable number inserted intron sequences (4–14) transmembrane helix (4–8) observed coding sequence Furthermore, phylogenetic analysis along with homologs from other species has clustered into six clades. Three clade V known regroup powdery dicots. Additionally, four core promoters predicted promoter region cis -regulatory elements related PM susceptibility.
